大鼠创伤性脑损伤后不同时间点的空间学习记忆能力及α7烟碱型乙酰胆碱受体表达与认知功能障碍的相关性

摘要:

目的 观察大鼠创伤性脑损伤(TBI)后不同时间点海马区及小胶质细胞上α7烟碱乙酰胆碱受体(α7nAChR)动态变化,探讨α7nAChR表达与认知功能障碍的相关性。方法 将48只SD大鼠分为假TBI(sham)组与TBI组,通过自由落体撞击实验制备TBI模型,各组在术后不同时间点(术后1、3、7、30 d),采用Morris水迷宫测试对大鼠空间学习记忆能力进行评估,采用免疫荧光染色标记小胶质细胞(MG)上的α7nAChR,采用Western印迹检测海马区域α7nAChR、白细胞介素(IL)-1β及IL-10蛋白表达,采用Pearson相关分析探究α7nAChR水平和水迷宫参数之间的相关性。结果 水迷宫结果显示,术后1 d, TBI组到达平台的潜伏期、距离、目标象限停留时间、穿越平台次数较sham组明显较差(P<0.05);术后3 d,两组水迷宫测试各项参数与sham组均无明显差异(P>0.05);术后7 d, TBI组潜伏期和距离较sham组明显较差(P<0.05);术后30 d, TBI组潜伏期、距离及穿越平台次数较sham组明显差(P<0.05)。免疫荧光染色显示,TBI后1、3、7 d在MG上表达的α7nAChR明显减少(P<).05)。TBI组在不同时间点大鼠海马区α7nAChR表达较sham组明显下降(P<0.05);在TBI后1、7、30 d促炎因子IL-1β蛋白表达水平明显增多,TBI后1、3 d抗炎因子IL-10蛋白表达明显增高(P<0.05)。α7nAChR表达与平均潜伏期呈负相关(r=-0.73,P<0.001),与目标象限停留时间呈正相关(r=0.44,P=0.03),与穿越平台次数呈正相关(r=0.65,P<0.001),α7nAChR表达与距离相关性不显著(r=-0.39,P=0.057)。结论 大鼠TBI后海马区及小胶质细胞上的α7nAChR表达显著减少,并与大鼠的学习和记忆功能障碍呈现相关性。
