目的 探讨隐丹参酮(CPT)调节Ras同源基因家族成员(Rho)A/Rho相关卷曲螺旋蛋白激酶(ROCK)信号通路对椎间盘退变大鼠髓核细胞凋亡的影响。方法 构建椎间盘退变大鼠模型,将造模后的大鼠随机分为模型组、CPT高、中、低剂量(45、15、5 mg/kg CPT)组及CPT高剂量(45 mg/kg CPT)+Rho激活剂(LPA,50μmol/L LPA)组,并以仅在皮肤做切口的大鼠为假手术组,磁共振成像检测大鼠椎间盘退变;甲苯胺蓝、原位末端标记法(TUNEL)染色分别检测髓核组织形态及细胞凋亡变化;酶联免疫吸附试验(ELISA)、实时荧光定量-聚合酶链反应(qRT-PCR)检测髓核组织中肿瘤坏死因子(TNF)-α及白细胞介素(IL)-1β水平及凋亡蛋白B细胞淋巴瘤(Bcl)-2、Bcl-2相关X蛋白(Bax)、胶原蛋白(HCB)Ⅱ表达;Western印迹检测髓核组织中RhoA、ROCK蛋白表达。结果 模型组较假手术组髓核细胞呈长梭形,有明显炎性浸润,Bcl-2、HCBⅡmRNA表达显著降低,Pfirrmann分级评分、细胞凋亡率、TNF-α和IL-1β水平、Bax mRNA、RhoA、ROCK蛋白表达显著增加(P<0.05);CPT高、中、低剂量组较模型组炎性浸润逐渐减少,细胞数量有所增加,Bcl-2、HCBⅡmRNA表达显著增加,Pfirrmann分级评分、细胞凋亡率、TNF-α和IL-1β水平、Bax mRNA、RhoA、ROCK蛋白表达显著降低,不同CPT存在组间显著差异(P<0.05);CPT高剂量+LPA组较CPT高剂量组炎性浸润有所增加,细胞数量有所减少,Bcl-2、HCBⅡmRNA表达显著降低,Pfirrmann分级评分、细胞凋亡率、TNF-α和IL-1β水平、Bax mRNA、RhoA、ROCK蛋白表达显著增加(P<0.05)。结论 CPT通过调节RhoA/ROCK信号通路改善椎间盘退变大鼠髓核细胞凋亡。
