目的 探讨吴茱萸碱(WZYJ)调节Toll样受体(TLR)4/核因子(NF)-κB/髓样分化因子(MyD)88信号通路对急性胰腺炎(AP)大鼠炎症反应的影响。方法 将SD大鼠分为Sham组、Model组、WZYJ低、高剂量(WZYJ-L、-H)组、WZYJ-H+TLR4/NF-κB/MyD88信号通路激活剂(LPS)组。酶联免疫吸附试验(ELISA)检测各组血清淀粉酶(AMY)、脂肪酶(LIP)、白细胞介素(IL)-10、IL-6、肿瘤坏死因子(TNF)-α水平;苏木素-伊红(HE)染色观察胰腺组织病理形态;TUNEL检测胰腺组织细胞凋亡;检测丙二醛(MDA)含量、超氧化物歧化酶(SOD)和过氧化氢酶(CAT)活性;Western印迹检测胰腺组织中裂解的半胱氨酸天冬氨酸蛋白酶(cleaved caspase)-3、TLR4、NF-κB、MyD88蛋白表达。结果 与Sham组相比,Model组胰腺组织结构明显被破坏,血清AMY、LIP、IL-6、TNF-α水平、胰腺组织细胞凋亡率、MDA含量及cleaved caspase-3、TLR4、NF-κB、MyD88蛋白表达显著升高,血清IL-10水平、胰腺组织SOD和CAT活性显著降低(P<0.05);与Model组相比,WZYJ-L、-H组胰腺组织形态有明显改善,血清AMY、LIP、IL-6、TNF-α水平、胰腺组织细胞凋亡率、MDA含量及cleaved caspase-3、TLR4、NF-κB、MyD88蛋白表达显著降低,血清IL-10水平、胰腺组织SOD和CAT活性显著升高(P<0.05);LPS可明显减弱WZYJ对AP大鼠的改善作用(P<0.05)。结论 WZYJ可减轻AP大鼠胰腺损伤、抑制炎症与氧化应激,可能与抑制TLR4/NF-κB/MyD88信号通路有关。
