目的 探讨睾丸相关高度保守的致癌长链非编码RNA(LncRNA THOR)的表达对胃癌细胞糖酵解、增殖和侵袭的影响及其对蛋白酪氨酸激酶(JAK)2/信号转导和转录激活因子(STAT)3信号的调控机制。方法 实时荧光定量-聚合酶链反应(qRT-PCR)检测胃癌组织和细胞中LncRNA THOR表达。以mimic-NC、mimic、mimic+AG490处理NCI-N87细胞;以Sh-NC、Sh和Sh+Colivelin处理AGS细胞;试剂盒检测葡萄糖的消耗量和乳酸生成量;Rh123检测细胞膜电位;5-乙炔基-2-脱氧尿嘧啶核苷染色(EdU)标记实验检测各组细胞增殖,吖啶橙(AO)染色检测各组细胞凋亡;Transwell小室检测各组细胞侵袭;小管形成实验检测细胞血管生成性能;免疫荧光检测神经型钙黏附蛋白(N-cad)和钙依赖细胞黏附分子(E-cad)表达。Western印迹检测各组细胞和小鼠肿瘤组织中JAK2、STAT3、p-JAK2、p-STAT3表达。结果 LncRNA THOR在胃癌组织和细胞系中的表达明显升高(P<0.05);升高LncRNA THOR的表达明显促进胃癌发展,发挥促癌效应,而抑制JAK2/SAT3的信号能部分明显逆转过表达LncRNA THOR对癌细胞的促癌效应(P<0.05);抑制LncRNA THOR的表达明显阻滞胃癌的发展,发挥抑癌效应,而激活JAK2/SAT3的信号能部分明显逆转下调LncRNA THOR对癌细胞的抑癌效应(P<0.05)。结论 LncRNA THOR在胃癌组织和细胞中的表达水平明显升高,LncRNA THOR通过调控JAK2/STAT3信号的传导,影响其磷酸化,进而影响胃癌细胞的糖酵解、增殖与侵袭性能。
