京尼平苷对慢性束缚应激诱导的老年小鼠抑郁样行为的影响

摘要:

目的 探讨京尼平苷(GP)对慢性束缚应激(CRS)诱导的老年小鼠抑郁样行为的影响。方法 20月龄雄性ICR品系小鼠随机分为对照组、对照+GP80(80 mg/kg GP)组、CRS组、CRS+盐酸氟西汀(Flu, 15 mg/kg Flu)组、CRS+GP40(40 mg/kg GP)组、CRS+GP80组各8只。除对照组和对照+GP80组外,其余各组每天束缚6 h,连续15 d,同时灌胃给药。采用蔗糖偏好试验、开场试验、悬尾试验和强迫游泳试验评估抑郁样行为;免疫组化法检测海马CA1区裂解胱天蛋白酶(Caspase)-3表达及B细胞淋巴瘤(Bcl)-2相关X蛋白(Bax)/Bcl-2比值;酶联免疫吸附试验(ELISA)检测海马白细胞介素(IL)-1β和肿瘤坏死因子(TNF)-α水平。结果 与对照组比较,CRS组蔗糖偏好率显著降低,开场穿越次数显著减少,悬尾、强迫游泳不动时间显著延长(P<0.05),海马CA1区裂解Caspase-3表达及Bax/Bcl-2比值和IL-1β、TNF-α水平显著升高(P<0.05)。与CRS组比较,GP(40、80 mg/kg)干预后上述指标均显著逆转,Flu(15 mg/kg)干预后上述指标可部分显著逆转(P<0.05)。结论 GP对CRS诱导的老年小鼠抑郁样行为具有改善作用,其机制可能与抑制海马神经元凋亡和减轻神经炎症有关。
