目的 探讨不同方法检测老年2型糖尿病患者血浆GLP-1水平的对比。方法 采集15例老年2型糖尿病患者空腹和餐后0.5、1.0、2.0、3.0 h各2份静脉血标本,分别加入丝氨酸蛋白酶抑制剂[4-(2-氨基乙基)苯磺酰氟盐酸盐(AEBSF)]和混合抑制剂[二肽基肽酶(DPP)-Ⅳ]的两种采血管中;使用流式荧光多因子(Luminex)技术和酶联免疫吸附试验(ELISA)分别测定其总GLP-1、活性GLP-1水平;比较两种抑制剂对老年2型糖尿病患者空腹和餐后GLP-1总水平及活性水平的影响;比较不同方法测定GLP-1水平的一致性。结果 使用AEBSF抑制剂或DPP-Ⅳ抑制剂时,Luminex与ELISA 2个检测方法间血浆总GLP-1水平在空腹、餐后1.0、3.0 h时无统计学差异(P>0.05),具有一致性;而活性GLP-1水平在空腹、餐后2.0、3.0 h时无统计学差异(P>0.05),具有一致性。使用Luminex和ELISA分别检测时,AEBSF和DPP-Ⅳ2种抑制剂于各个时间点血浆活性GLP-1水平均具有较好的一致性(P>0.05)。结论 在两种不同抑制剂下,血浆活性GLP-1水平仅偶尔于不同时间点具有一致性;两种不同检测方法下,血浆活性GLP-1水平在各时间点均具有较好一致性。
