目的 探究万古霉素对脂多糖(LPS)刺激肺癌细胞炎症反应、NOD样受体蛋白(NLRP)3/半胱氨酸蛋白酶(Caspase)-1信号的影响。方法 收集人肺癌A549细胞常规培养后将其分为对照组(经LPS诱导的A549细胞)、10、20、40μg/ml组(万古霉素10μg/ml+经LPS诱导的A549细胞)。采用酶联免疫吸附试验(ELISA)检测各组细胞中白细胞介素(IL)-1β、肿瘤坏死因子(TNF)-α、IL-6水平,采用Hoechst33342染色检测细胞凋亡形态,采用Transwell试验检测细胞侵袭;采用划痕试验检测细胞迁移;Western检测细胞中NLRP3、Caspase-1蛋白表达。结果 与对照组比较,10、20、40μg/ml组IL-1β、TNF-α、IL-6、细胞侵袭数量、NLRP3、Caspase-1蛋白、划痕愈合率明显降低,呈剂量依赖性(P<0.05);细胞凋亡形态显示,对照组可见明显细胞核固缩、团块现象;与对照组相比,10、20、40μg/ml组核固缩细胞数升高,其中40μg/ml组核固缩细胞数升高显著。结论 万古霉素对降低LPS诱导肺癌细胞引发的炎症反应和调节NLRP3/Caspase-1信号通路具有一定积极效用。
Abstract:Objective To investigate the effects of vancomycin on the inflammatory response and NOD-like receptor protein(NLRP)3/Caspase-1 signaling in lung cancer cells stimulated by lipopolysaccharide(LPS).Methods The human lung cancer A549 cells were cultured routinely and then divided into the control group(A549 cells induced by LPS),and the 10,20 and 40 μg/ml groups(10 μg/ml vancomycin+A549 cells induced by LPS).The enzyme-linked immunosorbent assay(ELISA) was used to detect levels of interleukin(IL)-1β,tumor necrosis factor(TNF)-α and IL-6 in the cells of each group.The apoptosis morphology of cells was detected by Hoechst33342 staining and the cell invasion was examined by Transwell assay.The scratch assay was used to detect cell migration, and Western blotting was employed to examine the protein expressions of NLRP3 and Caspase-1 in the cells.Results Compared with the control group, the levels of IL-1β,TNF-α,IL-6,the number of cell invasion, NLRP3,Caspase-1 protein and the scratch healing rate in the 10,20,and 40 μg/ml groups were significantly decreased, showing a dose-dependent pattern(P<0.05).The morphology of cell apoptosis showed that in the control group, obvious nuclear shrinkage and clumping phenomena were observed.Compared with the control group, the number of cells with nuclear pyknosis was increased in the 10,20,and 40 μg/ml groups, and the number of cells with nuclear pyknosis was significantly increased in the 40 μg/ml group.Conclusions Vancomycin has a positive effect on reducing LPS-induced inflammation in lung cancer cells and regulating NLRP3/Caspase-1 signaling pathway.
