瑞马唑仑调节Wnt/β-Catenin信号通路对喉鳞状细胞癌细胞恶性生物学行为的影响

摘要:

目的 研究瑞马唑仑通过调节Wnt/β-Catenin信号通路对喉鳞状细胞癌(LSCC)细胞恶性生物学行为的影响。方法 体外培养人喉鳞状细胞癌细胞AMC-HN-8,先用细胞计数(CCK-8)法检测瑞马唑仑对AMC-HN-8细胞的增殖能力影响,筛选合适的瑞马唑仑浓度,然后将AMC-HN-8细胞分为对照组、低浓度瑞马唑仑组(0.50μg/ml)、高浓度瑞马唑仑组(1.00μg/ml)、高浓度瑞马唑仑(1.00μg/ml)+Wnt/β-Catenin激活剂组(4 mmol/L Wnt/β-Catenin激活剂)。分组处理后,CCK-8法检测各组细胞活性;流式细胞仪检测细胞凋亡;Transwell小室检测细胞侵袭能力;创口愈合实验检测细胞迁移能力;Western印迹检测Wnt3a、β-Catenin、增殖细胞核抗原(PCNA)、B细胞淋巴瘤(Bcl)2蛋白表达。结果 与对照组比较,低、高浓度瑞马唑仑组细胞存活率降低、侵袭细胞数目减少、创口愈合率降低、Wnt3a、β-Catenin、PCNA、Bcl2、N-cadherin蛋白表达降低,细胞凋亡率、Cleaved-Caspase3、E-cadherin蛋白表达上升,差异有统计学意义(P<0.05);与高浓度瑞马唑仑组比较,高浓度瑞马唑仑+Wnt/β-Catenin激活剂组细胞存活率增加、细胞侵袭数目增多、创口愈合率升高、Wnt3a、β-Catenin、PCNA、Bcl2、N-cadherin蛋白表达升高,细胞凋亡率、Cleaved-Caspase3、E-cadherin蛋白表达降低,差异有统计学意义(P<0.05)。结论 瑞马唑仑可能通过对Wnt/β-Catenin信号通路的抑制作用,诱导LSCC细胞凋亡,并对细胞的增殖、迁移及侵袭能力产生抑制效应。
