冰茶洗剂通过Akt调控铁死亡-过氧化信号对糖尿病溃疡的修复机制

摘要:

目的 基于蛋白激酶B(Akt)调控铁死亡-过氧化信号探讨冰茶洗剂对糖尿病溃疡修复的生信分析及实验验证。方法 采用腹腔注射链脲佐菌素法建立糖尿病模型,并在大鼠背部脊椎骨两侧分别剪出直径为2cm的圆形全层皮肤缺损伤口。大鼠随机分为对照组、模型组、冰茶洗剂、冰茶洗剂+MK2206组(MK2206为Akt抑制剂,120 mg/kg);连续给药14 d;观察糖尿病溃疡创面愈合情况;采用苏木素-伊红(HE)染色观察大鼠胰腺组织病理改变;检测胰腺组织中铁死亡相关标志物铁、谷胱甘肽(GSH)、活性氧(ROS)、丙二醛(MDA)水平和Akt通路相关蛋白表达。结果 与对照组比较,模型组溃疡创面红肿明显,有少量血性渗出,创面愈合率显著偏低(P<0.01),血清中白细胞介素(IL)-1β、IL-6等炎性因子水平增加,肿瘤坏死因子(TNF)-α、碱性成纤维细胞生长因子(bFGF)、表皮细胞生长因子(EGF)及血管内皮生长因子(VEGF)含量明显降低,胰腺组织中GSH水平明显降低,铁、MDA水平及ROS含量明显升高,p-PI3K、p-Akt蛋白表达均明显降低(P<0.01);与模型组比较,冰茶洗剂组溃疡创面红肿明显改善,无渗出,创面愈合率明显增加,血清中IL-1β、IL-6水平明显下降,TNF-α、bFGF、EGF及VEGF含量明显升高,胰腺组织中GSH水平明显升高,铁、MDA水平及ROS含量明显降低,p-PI3K、p-Akt蛋白表达均明显升高(均P<0.01);冰茶洗剂+MK2206组中冰茶洗剂对糖尿病溃疡大鼠的保护作用被减弱。结论 冰茶洗剂可能通过激活AKT途径调节铁死亡相关因子以阻止铁死亡的发生,进而减轻脂质过氧化损伤,增强糖尿病溃疡大鼠的修复能力。
