目的 探讨甲基化CpG结合蛋白(MeCP)2对胶质瘤U87细胞的生物学影响。方法 应用siRNA干扰U87细胞MeCP2表达;采用实时荧光定量聚合酶链反应(qRT-PCR)法检测胶质瘤U87细胞中MeCP2含量;CCK-8法和集落形成法分析MeCP2对U87细胞增殖活力的影响;划痕实验和Transwell实验分别检测MeCP2对细胞迁移和侵袭能力的影响。结果 siRNA明显降低胶质瘤细胞U87中MeCP2表达(P<0.05);转染MeCP2 siRNA的U87细胞活力及增殖能力显著下降(P<0.05);MeCP2表达降低后U87细胞迁移和侵袭能力明显下降(P<0.05)。结论 MeCP2高表达能促进胶质瘤U87细胞增殖、迁移和侵袭。
